As others have said already...
The polarity of the speaker wires will make no difference to battery drain. The + & - designation is only an indication of the output from the amplifier to ensure that both/all speakers are in phase. I.e. the cones are pushing & pulling at the same time in the same direction. It does not not relate to +12V & Earth. Speakers are AC devices, not DC like most electrical items on a car.
If both rear speakers were connected the "wrong" way round compared to the fronts, or if one rear speaker was connected the "wrong" way and the other , the correct way, that would probably produce a kind of hollow or tinny tone to the music. Even more so if you have tweeters connected, which are out of phase or "wrong" way round.
It sounds more like the battery has drained while you've been working on the car with the interior lights on, maybe the fan going too... etc.
