It really depends on the condition of the sealer. I wouldn't go throwing the baby out with the bathwater, however, I also wouldn't leave a stone unturned.
You can get away with covering what's already present, as long as you ensure it's fully covered.
Your best bet is to apply a strip of sealant, then flatten it by using a ruler or even a wooden lolly stick.
This will ensure good coverage, with minimal waste.
Do it on a dry day (obviously), and if you decide to cover the roof, you'll probably need 2-3 tubes.
You can buy seam sealer or Tiger Seal in white, black or transparent flavours, so pick the colour that's most appropriate for your van.
