Great that it's constant; makes it easier to find. Nothing as bad as a internittent noise or fault.

Easy to check if the boot latch is the cause; unlatch the boot, (as if you were carrying a large load) and drive for a short distance with the boot open. (Use a bit of string if you don't want it right open, and even brace it against something (plastic recycling bin?) if you want to stop it flopping around; but you only need a couple of minutes to see if the noise is still active).

Seriously, the description of the noise matters; the stronger the thing being hit, and the heavier the vibrating bit, so the "thuddier" the noise is (more bass). if it's a nut in a tin-can, it can rattle faster, but you may find that shaking different parts of the car do not trigger the noise. Also try revving (up and down) the engine to generate it; the vibration and flexing of the engine and exhaust tends to show these things up quickly.

And, as described, I'd be surprised if it's the spare wheel; that won't move much on a straight road. If you're not having to do anything extreme, then the forces needed are not extreme (another good thing) and so you can find it by shaking things. Just try shaking the spare wheel; you'll not get it to move without quite a bit of force!
