After slicing my tendon putting one of these back last night (got it back and sliced it on the last bolt - I slipped and my finger slid along the back ledge of the frame - razor sharp on the under edge), I'll have a go at answering this...

I don't entirely know what that screw is for however, I suspect it to be to secure composite parts of the frame to each other. You could loosen it and put some sealant on but from the looks of things, I think the main problem is actually the outer seal. Ours was in tact but had become quite absorbant, letting water in all round. Yours looks to have broken a little and I think the water may be coming in at the seal and being channelled down that screw.

If the leak is occuring at that point with the roof open, then I shouldn't worry about it, as the channels will normally push the water to the drain hoses and not go near the screw. If it happens when closed then it will be due to one of the outer seals, either the roof to frame or the frame to window.
